The War Against Cancer - Can Beta Glucan Serve a Roll? Beta Glucan as a Primary Treatment for Cancer in Asia Since the first direct scientific study in 1963 clearly demonstrating the anti-tumor activity of beta-glucan, numerous animal, as well as human trials, have shown remarkable anti-tumor activity against a wide range of different tumors. In Japan, Beta Glucan is already licensed as an immunostimulant effective in cancer treatment. Two types of hot water extracts of Beta Glucan have successfully been used since the 1980s as traditional medicine for cancer therapy in Japan and China. One way in which the body fights cancer is a kind of lock-and-key action that identifies tumors as foreign and attracts various immune cells to engulf and destroy them. The basic approach of cancer immunotherapy (by using Beta Glucan) is to provide the body's immune system the necessary tools to facilitate this activity and destroy cancer cells. The human immune system is complex and is comprised of an innate and acquired immune system. The innate response is a non-specific response in the body by specific immune cells (macrophage, neutrophil and natural killer cells). Their function is to seek out and destroy "non-self" or foreign cells or antigens. The innate immune response is fast and effective against most pathogens, however, some foreign challenges are able to evade the innate response. The acquired or adaptive immune response takes over and involves the production of antibodies (humoral response) which recognize and bind to specific foreign antigens and facilitate the killing of these foreign cells, or the production of T-cells (cell mediated cytotoxicity) to kill specific foreign cells. Cancer cells are able to evade these innate and acquired immune responses. One therapeutic approach developed by scientists over the past 25 years is monoclonal antibodies. These are antibodies produced (cloned) externally and injected into the body to react with a specific foreign antigen on cancer cells. These antibodies bind to the cancer cell and "target" the cell as foreign, and allow the immune system to perform its function and kill the cancer. This therapy has been rapidly developing and currently there are more than 200 companies with over 300 monoclonal antibodies in various phases of drug development for a wide range of specific cancers. Whereas monoclonal antibodies are effective in binding to specific cancer and targeting them as foreign, they are not always effective in facilitating cancer cell killing. Transfer Point's Beta Glucan technology is effective in enhancing this immune response to more effectively kill the antibody targeted cancer cell. It performs this function because of a specific mechanism of action where the Beta Glucan binds to the carbohydrate binding region on the complement receptor 3 protein (CR3) on hte cell surface of the innate immune cells. This interaction between Beta Glucan and CR3 primes these immune cells and directs their enhanced killing action against tumor cells targeted as foreign, bearing a C3-antibody complex bound to the tumor cell. Beta glucan provides a "second signal" for the cells to activate a cancer killing immune response. This "one-two-punch" is a platform cancer technology in that it can be used effectively in combination with any monoclonal antibody that binds complement, and potentially a wide range of cancer vaccines. Preclinical research in breast cancer animal models have demonstrated the ability of injectable Beta Glucan alone to enhance the immune response and reduce tumor growth by 90% (figure 1). This occurs as some cancers trigger a humoral immune response where antibodies are produced that bind to the tumor and some of those antibodies fix complement, directing the immune response against the cancer. Some cancers, however, do not trigger an antibody response, limiting the effectiveness of Beta Glucan used alone. Monoclonal antibodies are used as a substitute for what nature has left out. In a preclinical study involving lyphoma, injectable Beta Glucan reduced tumor size by 30% and the monoclonal antibodies reduced it by 50% (figure 2). When these therapies were combined, however, tumor reduction reached nearly 80%. Similar results were also seen in a preclinical breast cancer study (figure 3). These results are highly promising. Ultimately, long-term remission is the desired outcome of cancer immunotherapy. One preclinical study on ling-term remission demonstrated that Beta Glucan combined with monoclonal antibodies was over 300 percent more effective than monoclonal antibody therapy alone. The significance of these discoveries is that Beta Glucan is a potential immuneotherapy adjuvant platform technology to enhance the effectiveness of the humoral repononse (natural, monoclonal antibody, or vaccine) against a wide range of cancers, including breast, lung, colon, lymphomas and melanoma. In addition to monoclonal antibody treatments, the company believes the technology will also be an adjuvant treatment with cancer vaccines in development against a wide range of cancer-types.
